چکیده

Direct manipulations of neuronal activity have been shown to induce changes in DNA methylation (DNAm), although little is known about the cellular signaling pathways involved. Using reduced representation bisulfite sequencing, we identify DNAm changes associated with moderate chronic depolarization in dissociated rat hippocampal cultures. Consistent with previous findings, these changes occurred primarily in the vicinity of loci implicated in neuronal function, being enriched in intergenic regions and underrepresented in CpG-rich promoter regulatory regions. We subsequently used 2 pharmacological interventions (nifedipine and FK-506) to test whether the identified changes depended on 2 interrelated signaling pathways known to mediate multiple forms of neuronal plasticity. Both pharmacological manipulations had notable effects on the extent and magnitude of depolarization-induced DNAm changes indicating that a high proportion of activity-induced changes are likely to be mediated by calcium entry through L-type CaV1 channels and/or downstream signaling via the calcium-dependent phosphatase calcineurin.

P 108: Evaluation of Calcineurin Role in Neuroinflmmation: Possible Targets for Early Detection and Treatment

Calcineurin (CaN) is a Ca2+/calmodulin (Ca2+/CaM)-dependent serine/threonine protein phosphatase expressed in most mammalian tissues but found at higher concentration in brain. In the last decade there have been a steadily increasing number of studies identifying neuronal CaN as a primary suspect in neuronal vulnerability, synapse loss, dendritic atrophy, synaptic dysfunction and neuroinflammat...

Calcineurin signaling mediates activity-dependent relocation of the axon initial segment.

The axon initial segment (AIS) is a specialized neuronal subcompartment located at the beginning of the axon that is crucially involved in both the generation of action potentials and the regulation of neuronal polarity. We recently showed that prolonged neuronal depolarization produces a distal shift of the entire AIS structure away from the cell body, a change associated with a decrease in ne...

Evaluation of epigenetic changes of liver tissue induced by oral administration of Titanium dioxide nanoparticles and possible protective role of Nigella Sativa oil, in adult male albino rats

Objective (s): Titanium dioxide nanoparticles (TiO2 NPs) have been recognized as biologically inert material and have been used in a multitude of applications. Nevertheless, the negative impact on the human health is not yet well understood. Aim of the work: The study attempted to evaluate the epigenetic changes of the genome, in the form of DNA methylation in liver tissue samples, resulting fr...

The role and importance of DNA methylation in spermatogenesis process

Background: DNA methylation is one of the epigenetic marks that are created by de novo DNA methylation and be maintained through cell division. This process is catalyzed by DNA methyltransferases. DNA methylation establishment in germ line is important, since they have the potential to regulate gene expression in offspring and improper DNA methylation patterns in germ lines has serious conseque...

Stable molecular changes during cell division without any change in the sequence of DNA molecules is known as epigenetic. Molecular mechanisms involved in this process, including histone modifications, methylation of DNA, protein complex and RNA antisense. Cancer genome changes happen through a combination of DNA hypermethylation, long-term epigenetic silencing with heterozygosis loss and genom...
